Zusammenfassung: | We studied variability in ribosomal RNA gene number in old-growth trees of Sitka spruce (Piceasitchensis (Bong.) Carr.). Total cellular DNA was extracted from two foliage samples taken from the ends of two major branches from each of 10 trees that were at least several hundred years old. Relative ribosomal RNA gene number was determined by probing slot blots with a cloned nuclear ribosomal gene from soybean. Through additional probings of the slot blots, these estimates were adjusted for residual variability in the amount of total genomic DNA blotted and the fraction of chloroplast DNA using multiple covariance analysis. Within trees, we found no indication of gene number differences among branches. Although small differences may have gone undetected, these results suggest that somatic variability of ribosomal RNA gene number is a minor source of genetic variability. |
