Zusammenfassung: | Efficient and rapid screening of recombinant proteins is an essential step for many applications in the areas of protein expression, function and structure research. Therefore, new ready‐to‐use 96‐well plates, His MultiTrap™ FF and His MultiTrap HP prepacked with Ni Sepharose™ 6 Fast Flow and Ni Sepharose High Performance, respectively have been developed. These plates are developed for screening and small‐scale high throughput purification of histidine‐tagged proteins.
Characteristics with the new prepacked 96‐well plates will be presented. First, un‐clarified sample can be directly applied to the wells. No significant differences in purity and recovery of purified protein loading un‐clarified compared to clarified sample were seen. Second, the prepacked format also gives high well‐to‐well and plate‐to‐plate reproducible results. Last the purification of histidine‐tagged proteins can easily be scaled up using HisTrap™ (1 and 5‐ml) column formats. Detergent solubility screening of membrane proteins, screening of soluble proteins and buffer matrix screening to optimize purification will be presented. Purifications of a membrane protein from the detergent solubility screening and a soluble protein using optimized buffer matrix were scaled up using HisTrap FF crude 1‐ml column. |
