Zusammenfassung:Diabetes is a condition of higher glucose content in blood which leads to several other complications like obesity, cardiac irregularities and artery problems. the objective of present study to evaluate Anti diabetic, anti obesity and antioxidant properties of J. Communis extract through different in-vitro assays. the extracts were evaluated for various in-vitro test to access the potential of J. Communis leaves against different enzymes and on the basis of their phytochemical analysis and prescreening with enzymes the methanolic extract was selected to test in adipogenesis in 3T3L1 mouse adipocytes and other enzyme inhibition assay like Amylase, pancreatic Lipase, alpha glucosidase and hydrogen peroxide assay were also performed to determine the anti diabetic activity. The cytotoxicity of extract was determined by the cell proliferation assay. The extract was evaluated for its anti obesity effect on adipocytes using 3T3L1 mouse adipocytes. J. Communis Extract showed the dose dependent reduction in triglycerides content in an assay. As per the results, it antagonizes the PPARγ through Ser112 phosphorylation via MAPK/ERK activation. In differentiated adipocytes culture. The extract treatment in differentiated adipocytes showed an increased (p = 0.0169) increased lipolysis as measured by the release of glycerol. J. Communis Extract does not exhibit cytotoxic effect on adipocytes. the study concludes that, J. Communis extract inhibits lipogenesis in adipocytes, antagonizes PPARγ and other lipogenic factors. It also accelerates triglyceride mobilization from the fat cells or enhances lipolysis. The other in-vitro enzyme inhibition was found with significant effect in treated groups against cellular and oxidative damage caused by free radicals.
