Zusammenfassung: | Objective: Laser scanning cytometry (LSC) is a technology that combines some features of flow cytometry (FCM), digital microscopy and image analysis. The goal of the work is to verify methodological procedures for processing formalin-fixed and paraffin-embedded tissue, which would be suitable for the analysis of archival material Methods: We processed tissues embedded in paraffin. 10 paraffin blocks with ovarian normal and carcinoma tissue. We prepared the monocellular suspension by enzymatic disintegration with 1 % pepsin. We used the method with heating and without heating. Results and Conclusions: By comparing the results of two methodical procedures without heating and with heating of the tissue section, we found that the methodical procedure without heating is a more suitable method for the preparation of monocellular suspension. |
