As2O3对慢性粒细胞白血病原代细胞BCR—ABL蛋白水平及信号转导的影响
目的:阐述As2O3对慢性粒细胞白血病(CML)原代细胞BCR-ABL及信号转导的影响。方法:采用免疫沉淀、蛋白酪氨酸激酶(protein tyrosine kinase,PTK)活性测定和Western印迹、实时定量PCR等方法。结果:1.0μmol/L和2.0μmol/L As2O3作用48h,CML患者骨髓单个核细胞BCR/ABL蛋白含量下调,0.5μmol/L时无明显改变。在0.5μmol/L As2O3作用下STAT1蛋白下调,1.5μmol/L和2.0μmol/L时下调更明显。P27蛋白无变化。As2O3作用60h,可使CML患者骨髓单个核细胞BCR-ABL蛋白PTK活性轻度下降。...
