Maternal educational attainment in pregnancy and epigenome-wide DNA methylation changes in the offspring from birth until adolescence

description | Maternal educational attainment (MEA) shapes offspring health through multiple potential pathways. Differential DNA methylation may provide a mechanistic understanding of these long-term associations. We aimed to quantify the associations of MEA with offspring DNA methylation levels at birth, in childhood and in adolescence. Using 37 studies from high-income countries, we performed meta-analysis of epigenome-wide association studies (EWAS) to quantify the associations of completed years of MEA at the time of pregnancy with offspring DNA methylation levels at birth (
n
= 9 881), in childhood (
n
= 2 017), and adolescence (
n
= 2 740), adjusting for relevant covariates. MEA was found to be associated with DNA methylation at 473 cytosine-phosphate-guanine sites at birth, one in childhood, and four in adolescence. We observed enrichment for findings from previous EWAS on maternal folate, vitamin-B
12
concentrations, maternal smoking, and pre-pregnancy BMI. The associations were directionally consistent with MEA being inversely associated with behaviours including smoking and BMI. Our findings form a bridge between socio-economic factors and biology and highlight potential pathways underlying effects of maternal education. The results broaden our understanding of bio-social associations linked to differential DNA methylation in multiple early stages of life. The data generated also offers an important resource to help a more precise understanding of the social determinants of health. |

<cop>London</cop><pub>Nature Publishing Group UK</pub><pmid>38052982</pmid><doi>10.1038/s41380-023-02331-5</doi><tpages>11</tpages><orcidid>https://orcid.org/0000-0001-7752-2585</orcidid><orcidid>https://orcid.org/0000-0002-7989-5833</orcidid><orcidid>https://orcid.org/0000-0002-4935-9298</orcidid><orcidid>https://orcid.org/0000-0001-6064-1588</orcidid><orcidid>https://orcid.org/0000-0002-3480-2031</orcidid><orcidid>https://orcid.org/0000-0003-0212-3509</orcidid><orcidid>https://orcid.org/0000-0003-4925-6325</orcidid><orcidid>https://orcid.org/0000-0001-5012-8983</orcidid><orcidid>https://orcid.org/0000-0001-6681-6983</orcidid><orcidid>https://orcid.org/0000-0002-9605-6337</orcidid><orcidid>https://orcid.org/0000-0002-1565-1629</orcidid><orcidid>https://orcid.org/0000-0002-4310-5297</orcidid><orcidid>https://orcid.org/0000-0003-1582-2747</orcidid><orcidid>https://orcid.org/0000-0002-6340-9300</orcidid><orcidid>https://orcid.org/0000-0003-2939-0041</orcidid><orcidid>https://orcid.org/0000-0003-4026-2964</orcidid><orcidid>https://orcid.org/0000-0002-1571-1468</orcidid><orcidid>https://orcid.org/0000-0002-9801-5774</orcidid><orcidid>https://orcid.org/0000-0002-8498-0020</orcidid><orcidid>https://orcid.org/0000-0001-8298-7272</orcidid><orcidid>https://orcid.org/0000-0001-9367-663X</orcidid><orcidid>https://orcid.org/0000-0003-1080-4339</orcidid><orcidid>https://orcid.org/0000-0003-4867-9465</orcidid><orcidid>https://orcid.org/0000-0003-4911-5290</orcidid><orcidid>https://orcid.org/0000-0002-6143-7172</orcidid><orcidid>https://orcid.org/0000-0002-2149-0630</orcidid><orcidid>https://orcid.org/0000-0001-7381-904X</orcidid><orcidid>https://orcid.org/0000-0001-7457-374X</orcidid><orcidid>https://orcid.org/0000-0003-1949-2298</orcidid><orcidid>https://orcid.org/0000-0002-7784-1612</orcidid><orcidid>https://orcid.org/0000-0002-8447-7871</orcidid><orcidid>https://orcid.org/0000-0002-6556-3635</orcidid><orcidid>https://orcid.org/0000-0002-8949-2587</orcidid><orcidid>https://orcid.org/0000-0002-2420-4118</orcidid><oa>free_for_read</oa></addata></record> |
