The nucleotide sequence of the 5′ end of papaya mosaic virus RNA: site of in vitro assembly initiation

The sequence of the first 138 nucleotides at the 5′ end of papaya mosaic virus (PMV) RNA spanning the initiation site for in vitro assembly has been determined by direct RNA sequencing. The first 35 nucleotides are free of uridine and rich in adenosine. Eight consecutive pentamer repeats (essentiall...

Zusammenfassung:The sequence of the first 138 nucleotides at the 5′ end of papaya mosaic virus (PMV) RNA spanning the initiation site for in vitro assembly has been determined by direct RNA sequencing. The first 35 nucleotides are free of uridine and rich in adenosine. Eight consecutive pentamer repeats (essentially GCAAA) are located immediately downstream of the cap site. The possible roles of these repeating adenosine-rich units in the nucleation of PMV assembly are discussed.
