Morpho-molecular evaluation for developmental competence of oocytes retrieved through transvaginal ovum pick-up from FSH-stimulated Tharparkar donor cows ( Bos indicus )

The study was conducted on indigenous Tharparkar cow ( ) to evaluate FSH stimulation on follicular attributes, oocyte recovery and morpho-molecular developmental competence parameters concerning oocyte quality. A total of 20 OPU sessions were performed, which included 10 sessions in each FSH stimula...

Findings on the size of follicles having ≥6 mm showed a significantly higher, however an opposite trend was observed in the case of smaller sized follicle (&lt;6 mm) between stimulated and non-stimulated respectively. The stimulated cows had a significantly higher number as well as the percentage of oocytes of Grade A, having a diameter ≥120 µm and BCB as compared to the non-stimulated cows. The relative mRNA expression profile of GDF9, BMP15, PCNA and BCL-2 genes was higher and BAX was lower in the FSH-stimulated cow. These results indicated that FSH stimulation before OPU in cows has a significant impact on follicle size, oocyte yield, recovery, and their quality with respect to COC's, diameter and BCB oocytes. Further, a significant increase in the relative mRNA expression levels of GDF9, BMP15 and PCNA genes in the FSH-stimulated group suggests that FSH plays a key role in modulating the expression of these important candidate genes and thus influencing oocyte quality. The higher mRNA expression of BCL-2 genes and concomitantly lower expression of BAX gene in FSH Stimulated cows indicates the protective role of these genes and preventing programmed cell death and thus promoting cell survival, quality and embryo development.</description><identifier>ISSN: 0967-1994</identifier><identifier>ISSN: 1469-8730</identifier><identifier>EISSN: 1469-8730</identifier><identifier>DOI: 10.1017/S0967199424000376</identifier><identifier>PMID: 39470002</identifier><language>eng</language><publisher>England</publisher><subject>Animals ; Bone Morphogenetic Protein 15 - genetics ; Bone Morphogenetic Protein 15 - pharmacology ; Cattle ; Female ; Fertilization in Vitro - methods ; Fertilization in Vitro - veterinary ; Follicle Stimulating Hormone - pharmacology ; Growth Differentiation Factor 9 - genetics ; Growth Differentiation Factor 9 - pharmacology ; Oocyte Retrieval - methods ; Oocyte Retrieval - veterinary ; Oocytes - cytology ; Oocytes - drug effects ; Oocytes - physiology ; Ovarian Follicle - drug effects ; Proliferating Cell Nuclear Antigen - genetics ; Proliferating Cell Nuclear Antigen - metabolism ; RNA, Messenger - genetics ; RNA, Messenger - metabolism</subject><ispartof>Zygote (Cambridge), 2024-10, Vol.32 (5), p.376-385</ispartof><lds50>peer_reviewed</lds50><oa>free_for_read</oa><woscitedreferencessubscribed>false</woscitedreferencessubscribed><cites>FETCH-LOGICAL-c226t-399fa40302e83d8a33036922344e00360e4251c02d99214cd9eff426afb1273b3</cites><orcidid>0000-0001-6153-0791</orcidid></display><links><openurl>$$Topenurl_article</openurl><openurlfulltext>$$Topenurlfull_article</openurlfulltext><thumbnail>$$Tsyndetics_thumb_exl</thumbnail><link.rule.ids>314,776,780,27901,27902</link.rule.ids><backlink>$$Uhttps://www.ncbi.nlm.nih.gov/pubmed/39470002$$D View this record in MEDLINE/PubMed$$Hfree_for_read</backlink></links><search><creatorcontrib>Donadkar, Manoj</creatorcontrib><creatorcontrib>Kumar, Brijesh</creatorcontrib><creatorcontrib>Singh, Sanjay Kumar</creatorcontrib><creatorcontrib>Chandra, Pradeep</creatorcontrib><creatorcontrib>Dangi, Pradeep</creatorcontrib><creatorcontrib>Gawai, Mohan</creatorcontrib><creatorcontrib>Jackson, Amala</creatorcontrib><creatorcontrib>Jasrotia, Nancy</creatorcontrib><creatorcontrib>Sharma, Shweta</creatorcontrib><creatorcontrib>Chouhan, Vikrant</creatorcontrib><creatorcontrib>Patra, M K</creatorcontrib><creatorcontrib>Khan, Meraj Haider</creatorcontrib><title>Morpho-molecular evaluation for developmental competence of oocytes retrieved through transvaginal ovum pick-up from FSH-stimulated Tharparkar donor cows ( Bos indicus )</title><title>Zygote (Cambridge)</title><addtitle>Zygote</addtitle><description>The study was conducted on indigenous Tharparkar cow ( ) to evaluate FSH stimulation on follicular attributes, oocyte recovery and morpho-molecular developmental competence parameters concerning oocyte quality.
