Rats were randomized to 5 groups of 8 rats each to: Control; Betel nut group; Varenicline at doses of 0.5, 1 and 2mg/kg along with betel nut. Conditioned Place Preference (CPP) model was used for studying the drug seeking behavior. The CPP score was compared between the groups. Results showed a significant increase in body weight of rats at the end of the study compared to baseline in betel nut and varenicline groups. Compared to betel nut group (656.25±55) there was significant decrease in CPP score in Varenicline group at 0.5mg/kg (97.5±48); 1mg/kg (18.75±20) and at dose of 2mg/kg (-60±6). The study demonstrates the benefits of varenicline in betelnut dependence. There was dose dependent decrease in betel nut seeking behavior with varenicline in rat model of drug dependence.
