description | Bisphenol A (BPA) is a chemical that is commonly applied in the manufacture of polycarbonate plastic and epoxy resins which are often used in food and beverage containers. Methylparaben (MeP) is commonly applied as food preservatives, products of personal cares, and medicines. This study was conducted to define the BPA and MeP effects on the formation of DNA-Adduct 8-Hydroxy-2’deoxyguanosine (8-OHdG) compounds. Analysis of 8-OHdG compounds was performed by applying a reverse-phase HPLC with a UV/Vis detector at the wavelength of 254nm. The optimum condition of HPLC was obtained with the mobile phase consisted of phosphate buffer solution and methanol at a ratio of 85:15 (v/v) and a flow rate of 1.2 mL/min. This research was conducted at pH 7.4 and temperature 37 °C with a variation of incubation time of 5 and 7 h. In this study, it was found that the presence of BPA and MeP in the mixtures produced an antagonistic effect to the formation of 8-OHdG compared to BPA and MeP in the separate reaction at the same condition of incubations. |
