Zusammenfassung: | Procedure: * Ethanolic solution of DPPH was prepared to a final concentration of 0.004% and served as negative control. * Ascorbic acid solution (5mg/ml) was used as positive control. * Similarly, different concentration of leaf extract was used and it was used as sample solution. * Reaction mixture was prepared by adding 100pl of leaf extract in varied range of concentration into 3ml of DPPH solution. * The reaction mixture was incubated for half an hour at room temperature under shaking condition and the absorbance was measured at 517 nm in UV-VIS Spectrophotometer. * The change in absorbance with respect to the control (containing DPPH only) was calculated as percentage scavenging activity12. .. Reducing power assay: * Reducing power was determined by the method prescribed14. * The sample in 1ml of methanol at various concentrations was mixed with a phosphate buffer (5 ml, 0.2 M, pH 6.6) and potassium ferric cyanide (5 ml, 1%), and the mixture was incubated at 50°C for 20 min. * Then add 5ml of trichloroaceticacid (10%) were added to the reaction mixture, which was then centrifuged at 3000 rpm for 10 min. * The upper layer of the solution (5 ml) was mixed with distilled water (5ml) and ferric chloride (1 ml, 1%), and the absorbance was measured at 700 nm. * A stronger absorbance will indicate increased reducing power. A mitochondrial enzyme in living cells, succinatedehydrogenase, cleaves the tetrazolium ring, converting the MTT to an insoluble purple formazan.
