Zusammenfassung:A highly sensitive and selective two‐photon fluorescence probe for catechol‐O‐methyltransferase (COMT) was developed via regioselective masking of one hydroxyl group of the substrates. The probe served as an efficient tool for biological sensing and bio‐imaging of COMT in complex biological systems, as well as for high‐throughput screening of COMT inhibitors. The strategy used in this study also sheds new light on the development of fluorescent probes for other conjugative enzymes.
