Effects of seed irradiation of ion beam (12C5+) on the M1 generation growth of netted melon (Cucumis melo L.)

description In order to elucidate the characteristics of ion beams as a mutagen for netted melon, the effect of seed irradiation with ion beams on the growth of M1 generation of netted melon was investigated. 1. Dry seeds were exposed to 220MeV carbon ions generated from AVF cyclotron (JAERI, Takasaki, Japan). The germination rate was not affected by the irradiation with the dose up to 200Gy, but the survival rate was decreased at around 80-100Gy and the most plants were dead at 160Gy. Thus, the median lethal dose (LD50/60d) was estimated to be between 110Gy and 120Gy. 2. Dry weight, pollen germination, fruit setting and seed fertility were decreased as the irradiation dose increased. All ovule numbers were not decreased by the irradiation up to 60Gy in the self-pollinating M2 seeds, but the seed fertility of ovules showed dose-dependent decrease, i.e., 58.2% at 20Gy and 39.3% at 40Gy. It was concluded that the irradiation dose necessary for the half decrease differed on each biological endpoint. 3. We will adopt 40Gy for the irradiation in which the endpoints related to seed production show half rates. 4. It was able to seem to apply ion beams as a mutagen for netted melon, because mutants such as chlorophyll mutation (albina) were obtained from the M2 generation.
