On utilise un piège optique pour translater une particule à travers le revêtement mince d'une surface optique plane. Le revêtement est de préférence hétérogène et le piège optique sert à déplacer la particule dans une succession de régions différentes du revêtement où se déroulent différents processus chimiques, biochimiques et biologiques tels que la synthèse et le séquençage d'oligonucléotides, la synthèse et le séquençage de peptides, la synthèse et le séquençage d'hydrates de carbone, la synthèse et le criblage de bibliothèques combinatoires, le séquençage usuel d'ADN (par exemple de Sanger ou de Maxam-Gilbert), ou le séquençage de molécules isolées d'ADN.
