According to the preparation method, an alkaline solution prepared from sodium hydroxide is used as a treating agent, and a sponge spicule is soaked and shaken, so as to achieve the purpose of reacting the alkaline solution with a silicon substance contained in the spicule, thereby generating a hollow channel in the spicule. Thus, the efficiency of transdermal drug delivery is improved, the drug utilization rate is improved, and more efficient, stable and safe transdermal absorption of a series of biomacromolecules such as drugs and cosmetic active components is promoted.
