La présente invention concerne une lignée cellulaire pluripotente humaine génétiquement modifiée où une séquence d'insertion comprenant une séquence nucléotidique d'un gène rapporteur fluorescent est insérée après un codon d'arrêt pour le gène α-MHC, le gène α-MHC et le gène rapporteur fluorescent étant ainsi co-exprimés. En manipulant génétiquement la lignée cellulaire pour qu'elle exprime le gène rapporteur fluorescent lors de l'expression du gène α-MHC, ce dernier étant un marqueur des cellules myocardiques, il est possible de suivre le processus de différenciation en observant le signal fluorescent lorsque la lignée cellulaire se différencie en cellules myocardiques. α-MHC 유전자의 정지코돈 뒤에 이어서, 형광리포터 유전자 염기서열을 포함하는 삽입서열이 삽입되어 α-MHC 유전자 및 형광리포터 유전자가 함께 발현되는 유전자 조작된 인간 전분화능 세포주를 제공한다. 심근세포 마커인 α-MHC 유전자의 발현에 의해 형광리포터 유전자가 발현되도록 유전자 조작을 하여 세포주를 심근세포로 분화시킬 때 형광 시그널을 관찰함으로써 분화 과정을 모니터링할 수 있다.
