L'invention concerne un procédé de détermination de la présence d'acide hyaluronique dans des échantillons complexes par clivage à l'aide de l'enzyme SpnHyl, puis réaction avec de la 3-méthyl-2-benzothiazolinone hydrazone (MBTH) pour former un produit coloré analysé par spectrophotométrie. Dans le cas d'une teneur élevée en glycosaminoglycanes, ceux-ci peuvent être séparés par précipitation avant le clivage, par exemple en utilisant du bromure de cétyltriméthylammonium (CTAB) dans un tampon acétate et un milieu salin.
