L'invention concerne des composés, des compositions et des méthodes de libération "intelligente" d'un agent thérapeutique, prophylactique ou diagnostique, telle que la libération d'insuline médiée par le glucose au moyen de dérivés d'insuline sensibles au glucose. Les dérivés d'insuline fixent le sérum-albumine ou s'agglomèrent in vivo. Les dérivés d'insuline se dissocient efficacement pour libérer l'insuline dans un état hyperglycémique, la complexation du glucose en élément sensible au glucose modifiant les propriétés du dérivé d'insuline menant à la dissociation.
