Cette invention se rapporte à un procédé servant à détecter la cytosine alkylée dans un ADN bicaténaire, en utilisant une ou plusieurs enzymes qui modifient de façon différentielle la cytosine alkylée et la cytosine. Au moins une zone de l'ADN est convertie en ADN monocaténaire et l'enzyme est amenée à réagir avec une zone cible de l'ADN monocaténaire. On détermine la présence ou le niveau de cytosine alkylée dans la zone cible en déterminant le niveau de modification enzymatique de la zone cible par l'enzyme.
