The present invention is related to a process for producing semapeptide, the process comprising the steps of, a) culturing a host cell comprising a nucleotide sequence encoding of Formula (II) under suitable conditions for expression, wherein, insoluble tag is a nucleotide sequence of Alanine-Valine; b) recovering semapeptide, wherein semapeptide amino acid sequence is Glu-Gly-Thr-Phe-Thr-Ser-Asp-Val-Ser-Ser-Tyr-Leu-Glu-Gly-Gln-Ala-Ala-Lys-Glu-Phe-Ile-Ala-Trp-Leu-Val-Arg-Gly-Arg-Gly.
