Zusammenfassung:FIELD: biotechnology, in particular preparation of human interferon for medical applications. ^ SUBSTANCE: Strain Escherichia coli BL21 (DE3)[pAYC-ET-(hIFN-alpha2b)-IacI) is constructed by gene engineering. Obtained strain is cultivated on in medium containing 10-20 g/l of casein hydrolyzate, streptomycin antibiotic in dose of 20-40 mg/l, 5-10 g/l of yeast extract and 0.5 tablet of vitamin-and mineral preparation "Centrum". Cultivation is carried out with introducing of 1,2-isopropyl-thio-beta-galactoside in dose of 0.1-0.2 g/l or lactose in dose of 6-8 g/l as interferon inducer into medium while maintaining of constant glucose concentration on level of 0.01-0.03 g/l and pH 6.7-6.8, and after inducing pH and glucose content is controlled by discrete introducing of glucose-containing additive in volume of 100 ml for 1 l of medium containing (mass %) acidic casein hydrolyzate 20-25; yeast extract 10-15; glucose 20-50, and MgSO4.7H2O 0.1-0.6.
