Zusammenfassung: | PURPOSE: Provided are the novel triarylimidazole derivatives and the pharmaceutically acceptable salts thereof and the compositions containing them to be used as the cyclooxygenase-2 inhibitor. CONSTITUTION: The novel triarylimidazole compound is represented by the chemical formula (1)where R1 is hydrogen, 2-,3-,4- or 3,5-halogen, 4-methyl or 3-fluoro-4- methoxy group, R2 is methyl, R3 is hydrogen, 4-halogen, 4-methyl, 4-benzyloxy, 4-hydroxy, 3,4-dimethoxy, 3-methoxy-4-hydroxy, 3- or 4-trifluoromethyl, 3-(3,5-dichloro)phenoxy, 4-nitro or phenoxy group. The synthetic process includes the steps of: making 1,3-dithiane compound from thioalkyl substituted benzaldehyde; alkylating the dithiane compound with chlorobenzaldehyde to make 2,2-disubstituted dithane; hydrolysing it to obtain benzoin derivative; converting the benzoin to triarylimidazole by reacting with ammonium acetate and benzaldehyde and finally oxidizing with oxone to make the compound as shown in the chemical formula 1. The conventional inhibitors are the 1,2-disubstituted imidazole derivatives and this invention is about 1,2,4-trisubstituted, structurally different novel compounds. |
