description PURPOSE:Human lymphoid cells which produce macrophage activation factor is cultured in a medium to obtain the titled RNA which mass-produces macrophage activation-factor (MAF) of high purity. CONSTITUTION:Human T lymphocytes are fused with human T lymphocyte tumor cells to obtain human lumphocyte cells such as a human T hybridoma producing MAF. The cells are inoculated in an enriched medium containing saccharides, aminoacids, animal serum and antigen in a concentration of 10 -10 cells/ml and cultured in a 5% CO2-95% air atmosphere at about 37 deg.C for 1-5 days. The cultured cells are crushed with a homogenizer or the like and extracted to recover the whole RNA. The whole RNA is fractionated with oligo dT cellulose column chromatography to obtains several kinds of RNA, e.g., corresponding to MAF; showing a sedimentation coefficient of 12-13.5s; having terminal adenyl acid structure; and coding MAF which is not neutralized with anti IFN-gamma antiserum.
