CONSTITUTION:A coupler substrate layer is laminated on a diazonium color reaction layer comprising a diazonium compound and a binder to form a multilayer analysis element. A molecular carbonic acid used as binder has a bivalent group derived from a compound having a carboxyl group and ethylene-like insaturated double bond as repeated unit and is preferably soluble in an organic solvent while non-soluble in water swelled by water. The high molecular carbonic acid meeting said requirements include acrylic acid copolymer, methacrylic acid copolymer, maleic acid copolymer and monoester maleic acid. This multilayer analysis element thus obtained is especially effective when the analyte is amylase.
