Zusammenfassung: | PROBLEM TO BE SOLVED: To obtain the subject liposome, having a high capturing efficiency of a plasmid, a high expression efficiency of a product with a cell and low toxicity to the cell and useful for gene therapy, etc., by using a specific amount of a specified lipid, etc., having a positive charge as a constituent ingredient. SOLUTION: This multilamellar liposome for embedding a plasmid therein comprises N-(α-trimethylammonioacetyl)-didodecyl-D-glutamate chloride(TMAG), dioctanoylphosphatidylcholine(DOPC) and dioleoylphosphatidylethanolamine(DOPE), which are positively charged lipids as constituent lipids. The multilamellar liposome is obtained by preparing a lipid thin membrane at 2:5:3 the molar ratio of the TMAG:DOPC:DOPE, adding a phosphoric acid buffer physiological saline solution containing the plasmid thereto and carrying out the vortex stirring. The resultant liposome has a high capturing efficiency of the plasmid, an ultrahigh expression efficiency of a product with a cell and ultralow toxicity to the cell and suitable for gene therapy, etc. |
