Zusammenfassung: | PURPOSE:To provide high efficiency in fractionation of blood plasma proteins by freezing and thawing a blood plasma protein-containing substance, subjecting the resultant substance to treatment with ethanol, anion exchanger, affinity chromatography, etc. CONSTITUTION:A blood plasma protein-containing substance is frozen, then thawed, filtered, etc., to provide a filtrate, etc., which are subsequently treated with ethanol in 5-10% concentration to recover fibrinogen, etc., from a precipitated fraction. The obtained supernatant is further treated with an anion exchanger to recover protopine, etc., from an adsorbed fraction. Unadsorbed fractions are then treated with affinity column chromatography using immobilized lysine to recover plasminogen from an adsorbed fraction. Unadsorbed fractions are further treated by the aforementioned chromatography with immobilized heparin to recover antithrombin-III from an adsorbed fraction. Unadsorbed fractions are subsequently treated with ethanol in 18-30% concentration to recover immunoglobulin, etc., from a precipitated fraction. The resultant supernatant is further treated with ethanol in 45% concentration to recover haptoglobin, etc., from a precipitated fraction and albumin from the supernatant. Thereby, the objective fractionation is carried out. |
