SOLUTION: Each of the primer couples are prepared by the following steps(a) to (c): (a) In an available DNA with a known sequence, a domain having any number of bases between 50 and 2,000 is selected, (b) the primer couple is synthesized, which is composed of a forward primer wherein 10-30 bases on the 5' terminal of the domain selected in the step(a) are joined to the 3' side of the forward primer for the multiplex PCR and a reverse primer wherein a complementary sequence of 10-30 bases on the 3' terminal of the domain selected in the step(a) is joined to the 3' side of the reverse primer, and (c) a PCR reaction is conducted using the primer couple synthesized in the step(b).
