Zusammenfassung:PROBLEM TO BE SOLVED: To obtain an S-hydroxynitrilelyase useful synthesizing an optically active S-cyanohydrin from a carbonyl compound with hydrocyanic acid in a good efficiency, high purity and high yield by culturing a specific yeast in a medium. SOLUTION: This method for producing an S-hydroxynitrilelyase is to culture an yeast belonging to the genus Pichia and transformed by a recombinant DNA obtained by incorporating a gene encoding the S-hydroxynitrilelyase (EC-4. 1. 2. 37) originated from a cassava, into an yeast chromosome-incorporating type expression vector. Further, it is preferable that the yeast chromosome- incorporating type expression vector consists of a DNA consisting of a base sequence of a AOX1 (alcohol oxidase) promoter originating from the yeast belonging to the genus Pichia, or a base sequence in which one or several bases are lost, substituted or added in the above base sequence, and containing the DNA maintaining the promoter activity.
