Zusammenfassung:	The present application discloses mutants of Taq enzyme, a method of preparation thereof and an application thereof. In the present application, the mutants of Taq enzyme comprises: an amino acid sequence having at least 70% identity to the amino acid sequence shown in SEQ ID NO:2; amino acid sequence is mutated at one or more sites selected from the group consisting of: D335, G499, E634, F769 and a combination thereof. The mutants of Taq enzyme provided in the present application have high amplification activity, yield more amplification products compared to wild-type Taq enzyme at the same number of PCR cycles, are both whole blood and high salt resistant, and do not lose 5'~3' exonuclease activity.
