The method is characterized by comprising the following steps of: filtering an image twice; extracting pink and purple pixel areas and combining the pink and purple pixel areas, preliminarily screening all connected domains of the obtained binary image, and extracting contours and positions of the reserved connected domains; traversing the connected domains, sequentially carrying out non-normalized sliding window square frame filtering, judging whether a new connected domain exists around a picture frame of the area, and if so, endowing a weight around the frame in a stepped manner; all obtained frames are scored after color judgment, the frame with the higher score preferentially becomes a pure bacterium scanning candidate area under the 100-time objective lens, and the frame position and the score are returned to a scanner for subsequent operation.
