Zusammenfassung: | The invention relates to a method for detecting influence of diet on oral absorption of rosuvastatin based on a PBPK model. The method comprises the following steps: collecting basic attribute parameters and clinical pharmacokinetic parameters of a rosuvastatin drug; establishing a rosuvastatin fasting and postprandial PBPK model; performing fitting verification on the model by using actually measured blood concentration data of oral administration of the healthy person on an empty stomach and after meal; and analyzing and exploring the influence of each factor on postprandial absorption of rosuvastatin by adopting parameter sensitivity. The PBPK model is helpful for exploring the possible mechanism that food causes reduced absorption of rosuvastatin, provides meaningful guidance for individualized treatment of the medicines, and provides reasonable diet suggestions for patients with hyperlipidemia to take BCSIII statins.
