According to the method, seismic data and logging data of a target exploration area are acquired; determining the total area of each helium-rich reservoir candidate zone, the granite distribution area, the uranium distribution area, the thorium distribution area, the uranium content, the thorium content, the fracture number, the type of the helium effective conduction system distribution area, the hydrocarbon source rock maturity, the seismic data type and the upper covering layer thickness in the target exploration area; determining a helium generation coefficient, a conduction coefficient and an aggregation coefficient of each helium-rich reservoir candidate zone based on each data; and determining a favorable zone of the helium-rich reservoir according to the helium source coefficient, the conduction coefficient and the trap coefficient.
