The preparation method comprises the following steps: preparing a non-noble metal doped surface enhanced Raman scattering substrate; aminated closed-loop rhodamine 6G is prepared; preparing mercury ion standard solutions with different concentrations, detecting by adopting the prepared surface-enhanced Raman scattering substrate, and drawing a standard curve according to peak values at the characteristic Raman shift positions of the obtained standard solutions with different concentrations and the concentrations of the mercury ion standard solutions; and detecting a pretreated water sample by using the obtained surface enhanced Raman scattering substrate, and comparing the obtained peak value at the characteristic Raman shift with the standard curve to obtain the concentration of mercury ions in the water sample.
