Zusammenfassung:	The invention relates to the technical field of biology, in particular to a preparation and genetic transformation method of cordyceps sobolifera protoplast, which specifically comprises the following steps: performing enzymolysis treatment on cordyceps sobolifera hyphae by using protoplast enzymatic hydrolysate, filtering the protoplast after enzymolysis to obtain filtrate, centrifuging the filtrate to remove supernatant, and washing precipitate by using STC buffer solution to obtain the cordyceps sobolifera protoplast, the method comprises the following steps: adding a target vector into protoplast, dropwise adding a PTC (Positive Temperature Coefficient) solution, resuscitating by using a regeneration liquid culture medium containing 0.7 M mannitol, and screening by using a resistance culture medium containing hygromycin to obtain a transformant. Compared with the prior art, the invention establishes a set of complete cordyceps sobolifera protoplast preparation, genetic transformation and screening method.
