The preparation method comprises the following steps: forming a PLGA-PEG copolymer from PLGA and NH2-PEG by using a chemical synthesis method, wrapping two mycobacterium tuberculosis fusion proteins ESAT6-Rv2626c and Mtb10.4-HspX (M4 for short) constructed in an experiment in the nanoparticles by using a multiple emulsion solvent evaporation method (W/O/W), modifying by using polydopamine and PolyI: C, and forming the PLGA-PEG-PolyI: C (NP for short) nano-particle tuberculosis subunit vaccine NP/M4 wrapping the mycobacterium tuberculosis fusion protein M4.
