The invention discloses a preparation method of acetified cotton pulp. The method comprises the following steps: selecting linters as a raw material, and carrying out the procedures of material preparation, cooking, pulping, bleaching and the like, wherein a cooking reagent comprises sodium hydroxide, hydrogen peroxide and an assistant in the cooking process; the assistant comprises modified glucose oxidase and at least one of modified fatty alcohol polyoxyethylene ether and an acrylamide copolymer; the amount of the modified glucose oxidase is 0.5%-3% of the weight of an oven dry material; the amount of the modified fatty alcohol polyoxyethylene ether is 0.5%-5% of weight of the oven dry material; and the amount of the acrylamide copolymer is 0.5%-3%. On one hand, the method for preparing the acetified cotton pulp is carried out at a relatively low temperature, a cellulose damage is small, the pulp is high in whiteness after being cooked and a black liquor is low in chemical oxygen demand (COD) and low in chromaticity; and on the other hand, the prepared acetified cotton pulp is high in yield, the R-10 content of the obtained acetified cotton pulp is greater than 98.2% and the PV value is greater than 600.
