Zusammenfassung:The present invention discloses a macrocyclic aryne compound containing a double helix structure, and a synthesis method thereof. The method comprises: under inert gas protection, heating Cu(OAc)2 and pyridine to a temperature of 50-70 DEG C, wherein per millimole of Cu(OAc)2 is added with 30-50 mL of pyridine; and slowly adding the pyridine solution of the intermediate g to the reaction system, carrying out a reaction for a certain time after completing the adding, cooling to a room temperature, carrying out pressure reducing pyridine removing, carrying out chromatography separation on the residue through a silica gel column, and further refining through HPLC to obtain the macrocyclic aryne compound containing the double helix structure, wherein per millimole of the intermediate g is added with 30-50 mL of pyridine. According to the present invention, the synthesis method of the macrocyclic molecule with the double alkyne bond being the coupling bridge and embedded with the double helix unit is used to carry out intermolecular matching on the oligomer so as to provide the suitable length of the chain containing the double helix unit and the suitable alkyne-terminated functional group, and then the intramolecular cyclization is performed to synthesize the macrocyclic compound, such that the steps of the synthesis method are less so as to improve the total synthesis efficiency.
