Zusammenfassung: | The invention provides a label-free thrombin detection method based on abasic sites. The detection method comprises the steps of synthesizing a double-stranded DNA by adopting a sequence as shown in SEQ ID NO: 1-2, introducing the abasic sites and a thrombin adapter in the double-stranded DNA simultaneously, and forming a compound with organic ligands for detecting the concentration of thrombin. According the double-stranded DNA, compared with an original double-stranded DNA, the length of the thrombin adapter at a 3' end is prolonged, target basic groups corresponding to AP sites are designed at prolongation parts, and the single strand of the DNA matched with the adapter chain part contains only ten basic groups (containing AP sites). The detection cost is reduced while the detection sensitivity is guaranteed. Moreover, the thrombin is detected by using the detection method provided by the invention, the enzyme specificity is high, the operation steps of labeling and cleaning in a traditional DNA probe are avoided, and the detection time is saved. |
