Zusammenfassung: | Determining whether a 1st protein (A) is capable of physically interacting with a 2nd protein (B) comprises providing a host cell which contains: (i) a reporter gene operably linked to a DNA-binding-protein recognition site; (ii) a 1st fusion gene which expresses (A), comprising a 1st protein covalently bonded to a binding moiety which is capable of specifically binding to the DNA-binding-protein recognition site; and (iii) a 2nd fusion gene which expresses (B), comprising a 2nd protein covalently bonded to a gene activating moiety and being conformationally-constrained, and measuring expression of the reporter gene as a measure of an interaction between (A) and (B). The same system is applied in: (i) detecting an interacting protein in a population of proteins; (ii) identifying a candidate interactor; and (iii) assaying an interaction between (A) and (B). |
