Zusammenfassung:The intricacy of multiple gene expressions frequently leads to various issues in metabolic engineering, from insufficient regulatory genetic parts for individual genes to suboptimal metabolic flux or imbalanced protein assembly, necessitating precise and predictable expression control strategies. We developed a synthetic module for accurate expression ratio by translational coupling for bacteria to handle the expression of multiple genes organized as a synthetic operon. The module uses translational coupling, one of the well-known polycistronic translation mechanisms, to prevent independent translation initiation from the downstream gene and allow its expression only when the upstream gene is translated. With the synthetic modules, we could precisely control the range of pathway gene expression, achieving up to a 7.7-fold change in valuable biochemical production. The data deposited here is about plasmids used for the validation of developed module and metabolite production (3-HP, P(3HB), and lycopene).
